Afinitor is used to treat adult patients with:

- advanced hormone receptor-positive breast cancer in postmenopausal women whose disease is no longer controlled by other treatments (so-called ‘non-steroidal aromatase inhibitors’). It is given with a type of medicine called exemestane, a steroidal aromatase inhibitor, which is used for hormonal anticancer treatment.

- advanced tumours called neuroendocrine tumours that originate in the stomach, intestines, lungs or pancreas. It is given if the tumours are inoperable and do not produce too much of specific hormones or other substances naturally secreted by the body.

- advanced kidney cancer (advanced renal cell carcinoma) when other treatments (so-called ‘VEGF-targeted therapy’) have not helped to stop your disease.

 

Contraindications:

Do not take Afinitor:

- if you are allergic to everolimus, to related substances such as sirolimus or temsirolimus, or to any of the other ingredients of this medicine.

 

Precautions:

Pregnancy, breast-feeding and fertility:

 

Pregnancy:

Afinitor may harm your unborn baby and is not recommended during pregnancy. Tell your doctor if you are pregnant or think you may be pregnant. Your doctor will discuss with you whether you should take this medicine during pregnancy.

 

Women who could become pregnant must use highly effective methods of contraception during treatment and for up to 8 weeks after stopping treatment. If, despite these measures, you suspect that you may be pregnant, ask your doctor for advice before continuing to take Afinitor.

 

Breastfeeding:

Afinitor may affect your breastfed newborn. You should not breastfeed during treatment and for 2 weeks after your last dose of Afinitor. Tell your doctor if you are breast-feeding.

 

Fertility in women:

Some women taking Afinitor have experienced a lack of menstrual periods (amenorrhea).

 

Afinitor may affect female fertility. Tell your doctor if you want to have children.

 

Fertility in men:

Afinitor may affect male fertility. Talk to your doctor if you want to have children.

 

Driving and using machines:

If you feel unusually tired (tiredness is a very common side effect), take special care when driving or using machines.

 

How to take it:

Always take this medicine exactly as your doctor or pharmacist has told you.

Talk to your doctor or pharmacist if you are not sure.

 

The recommended dose is 10 mg, taken once a day. Your doctor will tell you how many Afinitor tablets to take.

If you have liver problems, your doctor may start you on a lower dose of Afinitor (2.5, 5 or 7.5 mg per day).

If you experience certain side effects while taking Afinitor (see section 4), your doctor may reduce your dose or stop your treatment, for a short time or permanently.

Take Afinitor once a day, at about the same time each day, consistently with or without food.

Swallow the tablets whole with a glass of water. Do not chew or crush the tablets.

Composition:

- The active substance is everolimus.

- Each Afinitor 10 mg tablet contains 10 mg of everolimus.

- The other ingredients are butylhydroxytoluene, magnesium stearate, lactose monohydrate, hypromellose, crospovidone type A and anhydrous lactose.
